Cocaine use is known to induce an upregulation of dopamine transporters (DATs). To test whether cocaine use also increases [18F]fallypride binding, we studied seven chronic cocaine abusers with PET. Baseline evaluation was performed 1-2 days after the last use of cocaine. This study showed increased [18F]fallypride binding in five (71%) cocaine abusers. This upregulation was significant (P less than 0.01) in seven (100%) DAT binding and in six (86%) striatal regions. This upregulation persisted at the second PET series. The results show that cocaine use can increase [18F]fallypride binding, and that this increase is region-specific. This also confirms our earlier data that cocaine use can induce an upregulation of DATs.
The optimal age of learning to read is at least to age six, but before that the child must learn to form letters. Several discrete cortical areas are involved in reading, the temporal-occipital area being fundamental. An anatomically distinctly separate area has been identified in the superior temporal sulcus (STS) for semantic processing of language. This area contains the so-called word-form area, which may not be connected to reading but may be implicated in learning to read and other language abilities. A functional magnetic resonance imaging study was undertaken in 12 normal volunteers (N) and 12 primary school children (C), to establish the existence of this distinct area of activation in the STS. The children were between 7 and 9 years of age, and we excluded cases with known learning difficulties. Each subject performed two tasks on a magnetic resonance imaging scanner, a resting state and a lexical decision task. The activity in the two STS areas was analysed by paired and independent t-tests in each voxel and by calculating the averaged response from these two regions. The results indicated that children but not N demonstrate increased activation in the left part of the STS, a region which may play a role in reading. This area is distinct from the frontal cortex, which was activated by the task in both groups, but is consistent with a role for the STS in learning to read. d2c66b5586
